Controlled Processes

The BoneSTATION allows for a controlled and standardized bone fixation and decalcification process, optimizing parameters in all steps to achieve excellent results in the shortest possible time.

Fixation

Fixation
The fixation step can be performed using both the platforms.
  • The back platform (no heat) features a built-in magnetic stirrer that ensures fresh fixative solution is continuously exposed to the sample.
  • The front platform (heated) can reach temperatures up to 50° C and this speeds up the fixation step.
    It is equipped with a magnetic stirrer, which ensures the homogeneity of temperature throughout the solution, and an infrared sensor for automatic temperature control.

Decalcification

Declacification
The decalcification step can be performed using both platforms. However, the heated platform enables users to expedite the process. In addition to even heat distribution, the stirrer maintains a constant flow of fresh decalcifier onto the bone surface and removes the saturated solution from around the specimen, accelerating the decalcification process.

Productivity and Optimization

BoneSTATION enables outstanding productivity by concurrently run fixation and decalcification processes on its two platforms. Both processes are closely monitored, with continuous and detailed documentation of their parameters. The system doesn’t require cleaning cycles or maintenance, allowing for higher productivity and eliminating any downtime.
Recent reports have systematically investigated the effects of various fixation and decalcification agents on the quantity and quality of RNA and DNA recovered from bone biopsies. These reports concluded that the RNA and DNA yield and integrity are significantly reduced when strong acids (hydrochloric, nitric) are used in comparison with 12,5-14% EDTA and formic acid.

Analysis of the effect of various decalcification agents on the quality and quantity of nucleic acid (DNA and RNA) recovered from bone biopsies – Veena M. Singh et al. (2013)

For this reason, the use of either EDTA or formic acid as “soft” decalcifiers is preferred, as they are more suitable for molecular studies. The decalcification process is further expedited when EDTA is utilized within a pH range of 7.2-7.4. Based on this, Milestone has developed MoL-DECALCIFIER, an innovative pure EDTA solution, based on a stoichiometric mixture of basic and tribasic EDTA, able to reach this pH value without requiring an additional acid/base buffer. The combination of MoL-DECALCIFIER, temperature management, and the stirring function featured in the BoneSTATION significantly reduces decalcification times.

Safety

BoneSTATION can accommodate various types of fixatives/decalcifiers, including strong mineral acids such as HCl or HNO3, as all wetted surfaces are either glass or PTFE polymer. The system features closed vessels and fume condensation covers, and it must be installed under fume hood to further raise the safety during rack movement. The temperature of the reagent can be meticulously monitored using the temperature curve provided by the unit, ensuring controlled and safe processes.

Four acid-resistant rack configurations, each equipped with a built-in stirring bar, are available to accommodate bone specimens of varying sizes:

Full Documentation

With BoneSTATION, every factor influencing the fixation and decalcification process is documented and stored in a logbook, which can be transferred to a USB stick and printed for quality assurance and compliance purposes. This includes:
